    Evaluation of Forage Yield and Quality of Low-Lignin Alfalfa in Monoculture and Binary Mixtures in the Dark Brown Soil Zone of Saskatchewan

    Non-Peer ReviewedThe objective of this study was to compare low-lignin Hi-Gest® 360 alfalfa to a conventional legume in both monoculture and binary mixtures. Two varieties of alfalfa (monoculture) [Medicago sativa L. cv. AC Grazeland (Grazeland) and Hi-Gest 360 (Hi-Gest] or in mixture (binary) with AC Success hybrid bromegrass (HB) (Grazeland+HB and Hi-Gest+HB) were grown at a Dark Brown soil zone site (52o07′ N, 106o 38′ W) in Saskatchewan and harvested at 3 maturity stages (stage) of alfalfa [1 = 10% bloom; 2 = 40% bloom; and 3 = 100% bloom)] over 2 yr. Forage was harvested on June 21, June 25, and June 29 2018, and July 8, July 12, and July 16 2019. All plots were established in a randomized complete block design with four replications using forages as treatments. Plot size was 6.2 m × 1.2 m (7.44 m2) with 4 rows and total of 48 plots. Dry matter yield (DMY; 2-yr) and nutritive value (1-yr) of forages were determined. Results indicate that monoculture Hi-Gest alfalfa had greater (P 0.05) for forage quantity and quality. However, forages at maturity stage 3 had greater DMY (1058 vs. 783 kg), but lower CP (17.6 vs. 19.6%) and IVNDFD48 (37.1 vs. 42.3%) than those at maturity stage 1. The sugar content of forages linearly increased (P 0.05) observed between AC Grazeland+HB and Hi-Gest+HB mixtures for DMY or quality profiles. Monoculture had greater CP (18.5 vs. 12.0 %), ADFCP (2.5 vs. 1.5%), lignin (6.9 vs. 4.7%), fat (3.0 vs. 2.6%), RFV (174.8 vs. 91.8), NEm (1.6 vs. 1.3 Mcal/kg), NEg (0.96 vs. 0.77 Mcal/kg), but lower DMY (920 vs. 3343 kg/ha), ADF (27.4 vs. 35.9%), NDF (36.3 vs. 61.9%), starch (0.41 vs. 0.82%), and IVNDFD48 (39.2 vs. 50.7%), compared to binary mixtures. Results suggest that Hi-Gest alfalfa as a monoculture yielded less than AC Grazeland, however Hi-Gest monoculture had higher nutritive value than AC Grazeland, but was similar for yield and quality in mixture, suggesting Hi-Gest 360 alfalfa is a viable alternative legume for Dark Brown soil zone of Saskatchewan

    Predictions for oil slicks detected from satellite images using MyOcean forecasting data

    The pan-European capacity for the Ocean Monitoring and Forecasting (MyOcean) Marine Core Service, implementing the Global Monitoring for Environment and Security (GMES) objectives, targets the provision of ocean state observations from various platforms and analysis and forecasting products to assist, among other downscaling activities, the needs of the operational response to marine safety, particularly concerning oil spills. The MEDSLIK oil spill and trajectory prediction system makes use of the MyOcean regional and Cyprus Coastal Ocean Forecasting and Observing System (CYCOFOS) downscaled forecasting products for operational application in the Mediterranean and pre-operational use in the Black Sea. Advanced Synthetic Aperture Radar (ASAR) satellite remote sensing images from the European Space Agency (ESA) and European Maritime Safety Agency CleanSeaNet (EMSA-CSN) provide the means for routine monitoring of the southern European seas for the detection of illegal oil discharges. MEDSLIK offers various ways, to be described in this paper, of coupling the MyOcean forecasting data with ASAR images to provide both forecasts and hindcasts for such remotely observed oil slicks. The main concern will be the drift of the oil slick and also, in the case of the forecast mode, its diffusive spreading, although some attempt is also made to estimate the changes in the state of the oil. The successful link of the satellite-detected oil slicks with their operational predictions using the MyOcean products contributes to the operational response chain and the strengthening of maritime safety for accidental or illegal spills, in implementation of the Mediterranean Decision Support System for Marine Safety (MEDESS-4MS) regarding oil spills

    Efficacy of fatty acids dietary supplement in polyethylene glycol-induced mouse model of retinal degeneration

    Current knowledge of the benefits of nutrition supplements for eye pathologies is based largely on the use of appropriate animal models, together with defined dietary supplementation. Here, C57BL6 mice were subretinally injected with polyethylene glycol (PEG)-400, an established model of retinal degeneration with a dry age-related macular degeneration (AMD)-like phenotype, an eye pathology that lacks treatment. In response to PEG-400, markers of the complement system, angiogenesis,inflammation,gliosis,andmacrophageinfiltrationwereupregulatedinbothretinasand retinal pigment epithelium (RPE)/choroids, whereas dietary supplementation with a mixture based on fatty acids counteracted their upregulation. Major effects include a reduction of inflammation, in both retinas and RPE/choroids, and an inhibition of macrophage infiltration in the choroid, yet not in the retina, suggesting a targeted action through the choroidal vasculature. Histological analysis revealed a thinning of the outer nuclear layer (ONL), together with dysregulation of the epithelium layer in response to PEG-400. In addition, immunohistofluorescence demonstrated Müller cell gliosis and macrophage infiltration into subretinal tissues supporting the molecular findings. Reduced ONL thickness,gliosis,andmacrophageinfiltrationwerecounteractedbythedietsupplement. The present data suggest that fatty acids may represent a useful form of diet supplementation to prevent or limit the progression of dry AMD

    Low-Lignin Alfalfa Evaluated in the Black Soil Zone

    Non-Peer ReviewedA small-plot study was conducted at Lanigan (lat 51°51´N, long 105°02´W) SK, in the Black soil zone, to compare low-lignin Hi-Gest®360 alfalfa (Medicago sativa L.) to a conventional alfalfa (AC Grazeland, check cultivar). Each cultivar was seeded in August 2017 as both a monoculture and binary mixture (binary) with AC Success hybrid bromegrass (HB). Plants were sampled in 2019 at 3 different maturity stages of alfalfa (1 = 10% bloom; 2 = 40% bloom or commercial harvest stage; and 3 = 100% bloom). Forage was harvested 27 June, and 8 and 29 July, 2019 for stage 1, stage 2 (11 d after stage 1), and stage 3 (21 d after stage 2), respectively. Treatments were replicated 4 times, a total of 48 plots with plot size of 6.2 — 1.2 m. Forage — maturity stage interaction was not observed (P > 0.05) for all measured parameters. AC Grazeland and Hi-Gest exhibited similar (P > 0.05) DMY (2978 ± 405 kg/ha). Likewise, AC Grazeland+HB and Hi-Gest+HB did not vary (P = 0.62; 9120 ± 647 kg /ha) in DMY. There was little difference between Hi-Gest and AC Grazeland (P > 0.05) in nutrient profile. As alfalfa maturity advanced, DMY increased (P 0.05) were observed between AC Grazeland+HB and Hi-Gest+HB for nutrient profiles. The ADL concentration of Hi-Gest + HB was 106.3%, 93.7%, and 90.9% of (avg. 96.7%) AC Grazeland+HB, for the stage 1, stage 2, and stage 3, respectively. Hi-Gest alfalfa had greater (P 0.05) greater EDCP (8.2% more; 370 vs. 342 g/kg CP), but had similar EDDM (388.4 g/kg DM) and EDNDF (250 g/ kg NDF) relative to AC Grazeland+HB. In both mono- and binary systems, as the maturity advanced, EDDM, EDCP, and EDNDF declined (P Hi-Gest+HB > Hi-Gest > AC Grazeland. Compared to conventional alfalfa (AC Grazeland) that was harvested at the commercial harvest stage (stage 2; 40% bloom), Hi-Gest alfalfa harvested at stage 3 (100% bloom) had 169.23/haand169.23/ha and 143.57/ha greater market value in mono- and binary system, respectively. Preliminary results suggest that Hi-Gest alfalfa (in both monoculture and binary systems) did not differ in DMY and nutrient profiles, but was greater in rumen degradation potential relative to AC Grazeland, suggesting Hi-Gest 360 alfalfa is a viable alternative legume in both monoculture and binary systems for the Black soil zone of Saskatchewan

    Hindcast of oil-spill pollution during the Lebanon crisis in the Eastern Mediterranean, July–August 2006

    MOON (Mediterranean Operational Oceanography Network http://www.moon-oceanforecasting.eu) provides near-real-time information on oil-spill detection (ocean color and SAR) and predictions [ocean forecasts (MFS and CYCOFOS) and oil-spill predictions (MEDSLIK)]. We employ this system to study the Lebanese oil-pollution crisis in summer 2006 and thus to assist regional and local decision makers in Europe, regionally and locally. The MEDSLIK oil-spill predictions obtained using CYCOFOS high-resolution ocean fields are compared with those obtained using lower-resolution MFS hydrodynamics, and both are validated against satellite observations. The predicted beached oil distributions along the Lebanese and Syrian coasts are compared with in situ observations. The oil-spill predictions are able to simulate the northward movement of the oil spill, with the CYCOFOS predictions being in better agreement with satellite observations. Among the free MEDSLIK parameters tested in the sensitivity experiments, the drift factor appears to be the most relevant to improve the quality of the results.Published140–153JCR Journalrestricte

A MOON decision support system for oil spill monitoring and prediction comprising ocean colour satellite and SAR images, ocean current forecast (MFS-Mediterranean Forecasting System and CYCOFOS-CYprus Coastal Ocean Forecasting & Observing System) and the MEDSLIK oil spill model has been developed. The oil spill predictions obtained with MEDSLIK coupled to the CYCOFOS high-resolution ocean fields are compared with the oil spill predictions obtained using the lower resolution MFS hydrodynamics and both are validated against satellite observations. The predicted beached oil quatity along the Lebanese and Syrian coasts are compared with the in-situ observations.     Fast ultrasonic phased array inspection of complex geometries delivered through robotic manipulators and high speed data acquisition instrumentation

    Performance of modern robotic manipulators has enabled research and development of fast automated non-destructive testing (NDT) systems for complex geometries. This paper presents recent outcomes of work aimed at removing the bottleneck due to data acquisition rates, to fully exploit the scanning speed of modern 6-DoF manipulators. State of the art ultrasonic instrumentation has been integrated into a large robot cell to enable fast data acquisition, high scan resolutions and accurate positional encoding. A fibre optic connection between the ultrasonic instrument and the server computer enables data transfer rates up to 1.6GB/s. Multiple data collection methods are compared. Performance of the integrated system allows traditional ultrasonic phased array scanning as well as full matrix capture (FMC). In FMC configuration, linear scan speeds up to 156mm/s with 64 pulses per frame are achieved - this speed is only constrained by the acoustic wave propagation in the component. An 8x increase of the speed (up to 1.25m/s) can be achieved using multiple transmission elements, reaching the physical limits for acceptable acoustic alignment of transmission and reception paths.     Prehistory of Transit Searches

    Nowadays the more powerful method to detect extrasolar planets is the transit method. We review the planet transits which were anticipated, searched, and the first ones which were observed all through history. Indeed transits of planets in front of their star were first investigated and studied in the solar system. The first observations of sunspots were sometimes mistaken for transits of unknown planets. The first scientific observation and study of a transit in the solar system was the observation of Mercury transit by Pierre Gassendi in 1631. Because observations of Venus transits could give a way to determine the distance Sun-Earth, transits of Venus were overwhelmingly observed. Some objects which actually do not exist were searched by their hypothetical transits on the Sun, as some examples a Venus satellite and an infra-mercurial planet. We evoke the possibly first use of the hypothesis of an exoplanet transit to explain some periodic variations of the luminosity of a star, namely the star Algol, during the eighteen century. Then we review the predictions of detection of exoplanets by their transits, those predictions being sometimes ancient, and made by astronomers as well as popular science writers. However, these very interesting predictions were never published in peer-reviewed journals specialized in astronomical discoveries and results. A possible transit of the planet beta Pic b was observed in 1981. Shall we see another transit expected for the same planet during 2018? Today, some studies of transits which are connected to hypothetical extraterrestrial civilisations are published in astronomical refereed journals.     Acidic microenvironment plays a key role in human melanoma progression through a sustained exosome mediated transfer of clinically relevant metastatic molecules

    Background: Microenvironment cues involved in melanoma progression are largely unknown. Melanoma is highly influenced in its aggressive phenotype by the changes it determinates in its microenvironment, such as pH decrease, in turn influencing cancer cell invasiveness, progression and tissue remodelling through an abundant secretion of exosomes, dictating cancer strategy to the whole host. A role of exosomes in driving melanoma progression under microenvironmental acidity was never described. Methods: We studied four differently staged human melanoma lines, reflecting melanoma progression, under microenvironmental acidic pHs pressure ranging between pH 6.0-6.7. To estimate exosome secretion as a function of tumor stage and environmental pH, we applied a technique to generate native fluorescent exosomes characterized by vesicles integrity, size, density, markers expression, and quantifiable by direct FACS analysis. Functional roles of exosomes were tested in migration and invasion tests. Then we performed a comparative proteomic analysis of acid versus control exosomes to elucidate a specific signature involved in melanoma progression. Results: We found that metastatic melanoma secretes a higher exosome amount than primary melanoma, and that acidic pH increases exosome secretion when melanoma is in an intermediate stage, i.e. metastatic non-invasive. We were thus able to show that acidic pH influences the intercellular cross-talk mediated by exosomes. In fact when exposed to exosomes produced in an acidic medium, pH naïve melanoma cells acquire migratory and invasive capacities likely due to transfer of metastatic exosomal proteins, favoring cell motility and angiogenesis. A Prognoscan-based meta-analysis study of proteins enriched in acidic exosomes, identified 11 genes (HRAS, GANAB, CFL2, HSP90B1, HSP90AB1, GSN, HSPA1L, NRAS, HSPA5, TIMP3, HYOU1), significantly correlating with poor prognosis, whose high expression was in part confirmed in bioptic samples of lymph node metastases. Conclusions: A crucial step of melanoma progression does occur at melanoma intermediate -stage, when extracellular acidic pH induces an abundant release and intra-tumoral uptake of exosomes. Such exosomes are endowed with pro-invasive molecules of clinical relevance, which may provide a signature of melanoma advancement
